Since it has been two years I am going to assume you resolved this on your own. For myself and others that may later find this, I think I have discovered a way to resolve this.
The condition is that suddenly games will not boot and throw a 21H error. The memory check function will stop early at 128Kb, instead of the normal 640Kb.
It seems that this potentially has something to do with the bios, as completely removing the bios folder allows games to boot but with no text present and heavy glitches (obviously). Importantly, the issue is resolved by deleting the np2kai bios folder and replacing it with a known good copy. Inside this folder, I did notice that there was a cfg file, and this one file may possibly be the culprit of this issue. Try deleting that first, and see if it resolves the issue, otherwise just replace the entire folder. I don't know why but perhaps that file became corrupt.
I've been using the core to run many games for a long time now but suddenly the core won't boot any HDD image, not even a blank one and not even from floppy.
When booting from floppy it is complaining about interrupt 21H and when booting from HDD, it hangs after the line Copyright (C) 1981, 1994 Microsoft Corp. / NEC Corporation.
No keyboard buttons work at all.
The same NHD and floppy images boot just fine in the standalone NP2kai so I have no clue what happened. I haven't change any settings at all.
