How hard is it to devicify directed evolution? Seems one would definitely requires microfluidic for this purpose?
So far, I guess the step mainly involved 1) bioreactor 2) liquid handling robotics 3) optics assay
Seed the cells with mutant library (or mutator strains)
Choosing a target marker ( high yield, fast growing speed etc)
Push pressure (toxin antidote, phage infection, or auxotrophic mutant)
liquid handling part to screen ( high yield can be linked to GFP or better growth fitness OD)
Select and store
Sequencing to identify the changes.

Select and change is probably the hardest part, since we need to pipe the results back in as a new mutant library.
Off, the liquid handling is hard. However, like I point out in the paper, the phage pressured method allow you to have one uniform batch ( all strains are the same, the best man win) we can just store them to a reserving pot after each batch completed, then start with this and grow it up ( with plasmid of mutant library )
This one, not manual handling is required.

因为道长跟我说,要是能够用酵母人工合成大麻,会很酷炫。当时我就在想了想,其实抛去需要的基因组(synthetic gene circuits)我觉得技术难点对我们来说,应该是DIY bioreactor + directed evolution的一套系统。那么我以前帝国的一个老师,Mark Isalan 做过一个很酷的利用Bacteria Phage 来达到optimal protein interaction 的这样一组directed evolution系统。但是他们的系统是公司给做的,然后我又看到了用arduino bioreactor的一个paper (还没review),这个idea就油然而生
如何做一个电子宠物,让大家都简单的设定几个条件然后反应堆自行解答给你(我想要产大麻素最牛逼的酵母etc ) ?
