New S03 requested to assist in creation of P01 for 'total hydrolysable amino acids'.

Methods from metadata submission:

'Whatman glass fibre filter (GF/F, 25 mm diameter, 0.7 µm pore size), precombusted for 4 h at 450 °C were used for amino acid samples which were stored at -20 °C until analysis...For THAA, 2 ml of 12 M hydrochloric acid was added to 2 mL of sample in hydrolysis tubes, Oxygen-free nitrogen was bubbled through the solution for 1 minute, the tube was subsequently sealed tightly with a Teflon lined screw top and placed on a heat pad set to 110 °C for 6 hours. Once cooled, the sample extract was freeze-dried for at least 18 hours, Before analysis the sample was re-suspended in 2 ml HPLC grade water and 2 ml of OPA reagent added'

Proposed codnam: filtration, hydrolysis, reaction with orthophthalic acid

Definition: Physical separation of the solid phase from a volume of water or air before the phase of interest was treated with reagents to initiate a hydrolysis reaction followed by reaction with orthophthalic acid.

Review of chemical names - better to use ortho-phthalaldehyde than orthophthalic acid as originator refers to 'OPA reagent' in methodology. Similar S03 'S03115 - filtration, ortho-phthalaldehyde reaction' already in existence

Proposed codnam now: filtration, hydrolysis, reaction with ortho-phthalaldehyde

New S03 created: S03478 filtration, hydrolysis, reaction with ortho-phthalaldehyde Description: Physical separation of the solid phase from a volume of water or air before the phase of interest was treated with reagents to initiate a hydrolysis reaction followed by reaction with ortho-phthalaldehyde (OPA).
