As far as I can see, "AA AIO Tweaker" is a fork of the AA Phenotype Patcher, and both of them are licensed under the GNU GPL version 2. Given that it uses code from the original AA Phenotype Patcher, this software has to stay licensed under the GNU GPL, and the GPL forbids additional restrictions like "do not reupload". Unless you got explicit permission from Eselter, every contributor to the original repository, and every contributor to this repository, including everyone who created (merged) PRs to the repository, you cannot impose additional restrictions.
Everyone is free to download, re-upload and share copies of GPL software like AA AIO Tweaker, and the Readme shouldn't suggest otherwise. Filing a DMCA takedown request against copies of AA AIO Tweaker would be illegal as the software is under a license that explicitly allows hosting copies, unless I am missing something.
