Issues with the tool for extreme cases #9

Greetings, I've been using this wound healing size tool since it first came out and lately I have been noticing some issues with it when it comes to analyze images with extreme cases.

Scenario#1 Image link: https://www.dropbox.com/s/2wiacvwf3zwpblo/Case%231.tif?dl=0 This image represents a scenario where all cells are dead due a treatment and the scratch has, in a way, 100% percent area and a width that is same to the length of the image. However, which the tool is measuring it, it reports a good enough percent area (98.230%), but the width is way off (2.602 um).

Scenario#2 Image link: https://www.dropbox.com/s/jgawnkieuv8henl/Case%232.tif?dl=0 This image represents a different scenario where the scratch is separated by cells. The tool detects the opening at the bottom as the scratch with a 17% area and 18um width. While the percent area may be fine, I also think the width measurement is off for this case. I am also wondering if there is a why where I can define multiple ROIs as scratches and do the corresponding measurements.

The scale I used for both images are: Distance in pixels:1, Known distance: 0.43, Unit: um.

Hey Yun,

I apologize for my delayed response. After examining the images, you provided, I noticed some problems with the selected regions. My suggestion would be to manually choose the areas that you believe to be scratched using the latest version of the plugin for both cases. To avoid over-segmentation, the program only analyzes the largest ROI size. Therefore, in the second image, I would recommend creating an ROI that merges the two scratch regions before running the code.
