STORM Image Reconstruction Feature

Objective

The objective of this issue is to develop a robust STORM image reconstruction feature within our existing image preprocessing GUI. This feature will focus on high-resolution image reconstruction, specifically targeting super-resolution microscopy applications.

Requirements

• High-Resolution Imaging: The system must be capable of delivering images with precision in the 10-20 nm range.
• Multiple Sample Handling: The feature should handle multiple microscopy samples efficiently, ensuring consistent performance across various applications.
• Overcounting Analysis: Include mechanisms to analyze and adjust for overcounting in the imaging data.
• Frame-by-Frame Reconstruction: Implement the capability to reconstruct images frame by frame.

  Goals

• Develop an Image Localization Algorithm: To identify and localize points of interest in each frame with high precision.
• Create an Overcounting Adjustment Tool: This tool will analyze the count data to correct any biases or errors in the counting process.
• Integrate Frame-by-Frame Reconstruction: Ensure that each frame can be individually processed and reconstructed to form a cohesive high-resolution image.
• Ensure Compatibility and Performance: The feature must integrate seamlessly with a GUI and show real time reconstruction.

Challenges

• Algorithm Efficiency: Developing an efficient algorithm for localization that can process high-resolution data quickly and accurately.
• Data Handling: Managing large datasets typical of STORM imaging, ensuring the system remains responsive and stable.

Milestones

1. Research and Algorithm Design: Define and design the algorithms and parameters needed for localization and overcounting correction.
2. User Interface Development: Update the GUI to include controls and visualization tools for the new features.
3. Testing and Optimization: Testing across different data sets and further optimization based on test results.
4. Documentation and Deployment: Complete the documentation and prepare the system for deployment.

Expected Outcomes

• A fully functional image reconstruction module capable of producing high-resolution images from STORM data.
• Enhanced tools within the GUI for researchers to manipulate and analyze the reconstruction in real time.

In this moment the interface for STORM Image Processing has been finished as well as the single frame reconstruction which will then be used along with tracking of localized points to determine its movement and further final localization.

Next steps:

• Develop the tracking feature
• Save the results of tracking (this should contain by each frame some basic information of the estimated position for the molecule, intensity and on/off state as minimum.
