For stem-loop barcodes, allow loop to be barcoded/random too #16

Currently, stem-loop for barcodes are a fixed sequence, adding option o have this part of the barcode will add flexibility.

at same time should allow other regions to be a fixed sequence (instead of just polyA or random/barcode)
