mupen64plus-video-z64.so can only work when you also use mupen64plus-rsp-z64.so
You can only use mupen64plus-video-rice.so or mupen64plus-video-arachnoid.so or
mupen64plus-video-glide64.so when you use either mupen64plus-rsp-z64hlevideo.so
or mupen64plus-rsp-hle.so
m64py accidently selects mupen64plus-rsp-z64.so by default. So check that you
selected compatible rsp and video plugins
