This is a proximity ligation in situ assay. Also called P-LISA. PMID:15155907. The technique involves proximity probes and padlock probes. Proximity ligation is binders antibody with oligo nucleotide probes when brought into proximity given a ligation template these are ligated and can be amplified. this is very specific. padlock probe are ssDNA given ligation template can form circular ssDNA which can be amplified by a single primer rolling circle replication. This results in blobs made of 100's of concatemers, on using specific labelled oligos can be easily visulalised, spots correspond to single interactions. Plisa method soderberg in nature methods.
