How can I get a .nd movie (or other format) after doing eSRRF analysis? #10

Hello,

I am using NanoJ-eSRRF as pre-processing step to enhanced the movies for quantifying microtubule dynamics and, I cannot manage to get a movie showing microtubule growth after doing the analysis. Is it possible, if so how can I get a reconstructed video rather than just static images at the end?

Sorry for any inconvenience.

Hi, if you want to use only a subset of your total number of frames for SRRF processing you can set the # frames for SRRF to the frame window size you prefer (e.g. 50fr or 100) and it will reconstruct an image stack. image To increase the temporal sampling you can also activate the rolling window analysis. This will allow overlapping frame windows and will increase the number of reconstruction timepoints. This is also schematically shown in Extended Data Figure 7 in the publication : https://www.nature.com/articles/s41592-023-02057-w/figures/11 You can find some more info here: https://github.com/HenriquesLab/NanoJ-eSRRF/wiki/eSRRF---Main-reconstruction-parameters
