Is it possible to open the detected song in spotify instead of google assistant when the now playing banner on the lock screen is tapped?

No, sorry. The mod doesn't have the capability to look up singular songs so it would have to load the database every time and that would make the action extremely slow. Much quicker to open Now Playing and go from there.
