  1. Set up click reaction by adding sodium ascorbate (47.9 mg, 0.242 mmol, 0.88 equiv) and CuSO4.5H2O (26.7 mg, 0.107 mmol, 0.38 equiv.) to a 3-neck flask and evacuating/filling with N2 five times.

NB - equivalents based off cyclam KBS10

  1. Added KBS10 (132 mg, 0.276 mmol) by solution transfer in 2 mL THF
  2. Added KBS16 (112 mg, 0.466 mmol, 1.7 equiv.) by solution transfer in 2 mL THF
  3. Added 3 mL water and stirred at rt from 4.30 pm

  1. TLC at 2 pm indicated completion (5:95 MeOH:DCM). Reaction was olive green with two layers forming.
  1. THF was removed by rotovapping. The mixture turned a reddish brown colour and started to form a gummy solid.
  2. The reaction was diluted with ammonium chloride buffer (10 mL) to remove excess Cu, and extracted with DCM (3 x 15 mL). During extraction, the organic layer was clearly reddish brown while the aqueous remained olive green.
  1. Organic extracts were dried over MgSO4 and rotovapped down. The initial solution was amber coloured but upon rotovapping the mixture went back to a greenish colour and formed a foam.

Vial (empty) = 15.5737 g Vial (full) = 15.7910 g Mass = 217.3 mg

Peaks: 959.44 [M+H], 981.42 [M + Na].

Evidence of product, no evidence of mono-coupled product (mw = 717). Very minor peak at 680 - uncertain as to provenance.

Columned crude product with 8:92 MeOH/DCM (9 CV) and isolated fractions 3 and 4.

1H NMR (300 MHz, CDCl3) δ 5.30, 4.27 (4H), 4.19 (4H), 4.18 (10H), 3.69 (4H), 3.28 (8H), 2.55 (4H), 2.42 (4H), 1.40 (18H), 1.26 (4H).

Cleaned sample for characterisation (hexane washed). Finally clean - can submit for hi-res NMR, and get IR.
