Want to check HI LO results for the following compounds:

740, 741, 744, 746, 748, 752, 755, 758, 768

Inoculated 2 tubes using DEY plate subbed 1/10 (mm55 2018).

Also inoculated 2 tubes using CBS247.48/8 plates subbed 3/10, as they seemed to have grown well. Used half a plate each but did drop some fragments.

~20 mL media total per tube

Tube A: poured off til 5 mL

12.5 mL rpmi + 3 mL = 81.8

• 1 mL = 72.3

Used for plate 2

Tube B: poured off til 5 mL 12.5 mL rpmi + 5 mL = 86.9

• 2.5 mL = 81.9 Removed 5 mL and added 1.25 suspension = 68.2

CBS tube B is possibly contaminated but may also be from agar plate. Use tube B anyway as tube A had very little hyphal growth - I did also forget to add media before sonicating which may have caused the issue.

Tube B transmissions were 68 and 69% for plates 1 and 2 respectively.

Made up mm55 plates as planned.

Discarded contaminated CBS plates as control had clearly not worked.

Added MTS (7.5 uL) at ~10.25 and allowed to incubate 20 minutes before spinning down and transferring to a flat bottomed plate.
