Some quick calculations indicate that the LIP-X1 at 1x should be pretty reliable with this, but images don't come out good even on a fairly well tuned system. The following tweaks would be ideal:
Correct camera angle vs stage alignment in software. We are always going to have a small offset / annoying to mechanically fix
Crop images instead of pasting edge on edge. This will minimize edge lighting issues
Consider revisiting flat field correction. Its now much easier to see the effect vs the more opaque cloud stitching workflow that automatically corrects a lot of issues
Verify 1x is actually very close to 1/20 of 20x (the scale calibration objective)
