Interaction type / physical / direct / 1:1 Direct physical interaction between two molecules that is characterized by a fully reversible and single (constant) association and dissociation rates. (Langmuir) other Interaction that can not be described by a 1:1 binding model. For example, one of the interactant has two or more binding sites (antibody), the interactant is not pure (for example because of various post translational modifications) or when an interaction involves a conformational change of the complex resulting in a conformation dependent dissociation rate.

Kinetic parameters are important to understand the biological impact, i.e. what happens with a biological process when a concentration of one or more of the interacting proteins changes. If the ambition of the interaction data base is to understand (model) biological processes in living organisms (with rapid fluctuating protein concentration levels), information on kinetic parameters are a must. Interactions are strongly dependent on environment conditions like temperature, pH, and buffer composition. The temperature at which the interaction is determined is often well defined and a temperature term would allow easily refinement of the information retrieved from a database. To model influences of changing protein concentrations in biological processes, it is also important to have a “quality level” on the interaction parameters (1:1 or other).
