Use both positive and negative versions.

Note: A binomial test can be performed to test for the significance of a co-occurrence pattern. a=occurrence of species a b=occurrence of species b ab=co-occurrence of a and b N = number of observations Nab = number of co-occurrences P(P(ab)=0.5) = binomial(Nab,N,p=0.5)
