Add philosophical point about cells as replicates #10

Mention how it's conceptually inappropriate to treat cells from different functional subpopulations as replicates in a standard DE analysis.

... but also add a mention of how that would be useful for identifying marker genes.

Scratch the marker gene point, actually - it's a lot more complex than I first considered. However, it may be desirable to move the section about modelling cell-to-cell variability into the main text.
