From that it appears logical that increasing fu_Hep will result in a lower Clint, if t1/2 is kept constant as it is together with the t1/2 in the denominator.

Dear all,
I am trying to use the IVIVE option in PK-Sim which allows you to input the T/2 in hepatocytes and calculate the intrinsic clearance. Is there a reference for the equation used (see screenshot below)? In this equation there is the fraction unbound in the essay, which in this case would be Fu_Hep and I am having problems to understand why increasing the Fu_Hep for the same T/2 we see a decrease in the clearance. (Since from my point of view: higher Fu_Hep= more drug available in the essay, so for the same T/2 this would mean higher clearance...) Any help is highly appreciated! Thanks, Donato
