The easiest way to add additional variability to a GFR process is to add variability to the GFR fraction
in a population simulation.
A total variabilility of a GFR process will then be a combination of variabilities of GFR specific, GFR fraction and Kidney Volume in a population simulation.
P.S. In order to be able to add additional variability to the GFR spec on a population level, we would need new GFR scale factor (similiar to Plasma Protein Scale factor for fu)

Moving that to PKSim so that we can discuss if it makes sense.
I have the same question regarding how to set up the boundary condition for GFR in PK-Sim. For instance, the healthy subjects have GFR values greater than 90 ml/min/1.73m2 and the subjects with mild renal impairment have GFR values >60-<89 ml/min/1.73m2. The individual can be defined with the respective kidney volume, GFR specific, and renal blood flow for subjects with different stages of renal impairment. Then RI populations are created. According to your suggestion, I can change the variability of GFR fraction. But as you said, “A total variability of a GFR process will then be a combination of variabilities of GFR specific, GFR fraction and Kidney Volume in a population simulation”. My question is how to set up the boundary condition for GFR in PK-Sim and how to make sure the generated population with the GFR values within that boundary. In addition, where can I find the GFR output for individual subject in a population simulation.
Thanks!

Dear PK-Sim community, I am currently creating population simulations for my project. The populations described in the clinical study used for modeling are divided into groups based on their renal function (healthy, mild renal impairment, moderate renal impairment…). To recreate the reported populations, I want to specify a range for the GFR specific in my population simulations. Unfortunately, I am not able to access this parameter in the simulation in the tab “User Defined Variability”. Is there a way to access this parameter or any other parameter related to renal function to specify a range for the GFR in my population simulations?
Thank you for your support!
