Open mattodd opened 4 years ago

On the call: @fantasy121, @kym834, @wwjvdsande, @MKonings, @Wilson-Lm, @mattodd, @MFernflower, @dmitrij176, @bendndi and @MOUSEY007

@alintheopen could not attend.

Recording of the meeting Open Source Mycetoma Community Monthly Meeting #4 27th Oct 2020 - YouTube.

  1. Team USYD update - @fantasy121 has been working on the synthesis of a number of compounds. Still waiting on a few reagents to arrive. Data for these will be added to the share document for the manuscript. No update from @kym834.

    • [ ] ACTION ITEM: @fantasy121 to add data into master doc.
  2. Team UCL update - 3 new compounds made, still working on characterization. P4-A-0080 has been received from Sevenoaks. Still needs purification and then characterization. @dmitrij176 working on this.

    • [ ] ACTION ITEM: @dmitrij176 to add data into master doc when finished.
  3. Unique MYOS codes for compounds - once compounds have been made and characterized need to be given a MYOS code to uniquely identify. Compounds that are have not yet been made or are just an idea will not be given MYOS code. How to code these has been discussed here

    • [ ] ACTION ITEM: @dmitrij176 to assign codes to the master list and add in a new column that has the historical identifier so that we can follow it back if need be.
  4. Stability of molecules in solution - Having some issues with result reproducibility for some compounds. With a lag phase between /in vitro/ and /in vivo/ testing of molecules it would be good to know if they are stable over this time. Compounds are stored in DMSO, 10 uM, -20 °C in a sealed container. @dmitrij176 has experienced degradation of DM11-2 over time. Solutions: store in DMSO-d6 under the same conditions and run NMR every month to assess stability and/or

@Wilson-Lm noticed that HPD009-1 sticks to plastic. May want to consider which plastic is being used, polypropylene or polystyrene as these have different binding affinities to the compounds ( thank you @MOUSEY007 ) - during synthesis very limited contact with plastic. Something to consider as we progress further into in animal studies.

  1. Team Erasmus update - presentation slides below. First /in vivo/ experiment for 14 compounds with MIC > 4 uM. Four compounds improved larvae survival at 20 uM per larvae HPD17-2F3, HOD30-1, HPD0033-3P3 and DM7-1. Still need to be repeated to confirm. Running 2nd and 3rd /in vivo/ experiments now. @wilson-Lm will update the results and added to GitHub.
