  1. Discussion on the outstanding items from previous meeting • The first action item was to look up compunds in the Epichem library which could fill in the holes of the SAR. The link of the library is found here. @fantasy121 and @dmitrij176 will both go to the Epichem library and create a list together. These compounds would then be requested and shipped for in vitro screening. • @bebi78 had sent an e-mail to @bendndi whether any samples of benzoxazoles (structurally related to series 5) might be available from DNDi's other projects. Seventeen compounds will be sent. There are new compounds incoming. It is in the repo of series5. @wwjvdsande just added the shipping address in the repo. • @kym834 to update on series 2 work with SSP and high school students and double check there is no doubling of compounds synthesis from different participating high schools (Australia vs Seven Oaks for eg). Compounds were sent.

  2. Data from the different labs • USYD update: USYD was absent this meeting. So no updates here. • UCL update: one last fenarimol was synthesized. Three ketoximes are made. As a back-up option there was one molecule DM62-1. DM61-1, DM64-1, DM72-1 and DM76-1 were not stable, they detoriated. • Erasmus update: @MaJingyi had screened several compounds twice in the galleria model. The third repeat is still going on. Together with the compounds the following controls are included: o Non-infected control (aquadest) o Infected control. This is the negative control. Larvae treated with solvent only will almost all die within the 10 day observation periode. o AMB control (amphotericin B control). This control is used as a positive control. AMB always prolongs larval survival. For series 1 the following compounds were screened: MYOS_00166 (HPD045+1P3Cryst); MYOS_00174 (HPD044-1); MYOS_00204 (DM54-1) and MYOS_00206 (DM56-1). Unfortunately none of these compounds prolonged larval survival. For series 6 MYOS_00193 (DM34-1) was screened. This compound did not prolong larval survival. For the natural compounds, MYOS_00232 (naphthazarin) was tested. This compound did not prolong larval survival. Determining the therapeutic efficacy for the other compounds is ongoing.

  3. Manuscript • It is important to work further on the manuscript. To finalize the work on series 1. • @fantasy121 and @dmitrij176 to update characterization data, for the manuscript and for the in vivo compounds. The main data needs to be polished. Dmitrij has half of the NMRs done. However for the first half they were run on a different machine. In worst case scenario some needs to be rerun. This part needs to be finished to be able to finish the paper. We need the full write up for the paper to be ready to go when we finished. Because it is an open science paper the NMR data needs to be uploaded so that people can redo this when they want. The SI document needs to be updated. All compounds need to have the raw NMR data ready.

  4. Any other businesses? Matt is going to have a talk of a head-master conference to get some more schools interested in MycetOS. Wilson will use the current version of the fenarimol paper for this thesis.

  5. Action Items • @mattodd @wwjvdsande @bendndi to coordinate submission to UNESCO call by July 15th. Since it is today 28 June, we need to work on this next week at the latest. @mattodd will take the lead and @wwjvdsande will provide some biological data when needed.

Please could people also, if they have been involved in signfiicant discussions over the past month (e.g. on paper writing or compound synthesis or OpenSourceMycetoma/Adjuvant-Screen#1 or OpenSourceMycetoma/Adjuvant-Screen#2) to please come with a short summary of those activities.

Hi Mat, I’m unable to attend the meeting. Could someone else please chair this session and I’ll chair the next meeting we have. Thanks.

I will write an update of my progress here by the end of the week.

Hi @OpenSourceMycetoma/corecontrib - yes, if Sydney are not joining then it makes sense to move the time. I could do 12:30 London time today, which is 13:30 Euro time? Any preferences from anyone?

@OpenSourceMycetoma/corecontrib I have a second meeting starting at 14:00 Euro time. So 13:30 is a bit short may be. However I can move to 12:00 euro time or 15:00 euro time.
