• [x] Decide on the monthly MycetOS meeting schedule for the upcoming months. In the meeting: it was decided to move to a time that is the same each month: 12 noon UK, 1 EU and 9 Sydney. It was also decided that
• [x] @mattodd would generate a back-up Zoom link should USyd not be able to attend, and that
• [x] @wwjvdsande would generate a Teams link as option number 3.
• [x] @wwjvdsande to include these links in a new Outlook invitation for the next 12 months worth of meetings. The meetings are on the 4th Tuesday of each month
• [x] @fantasy121 or @kym834 (whoever made them) to cancel the Outlook invites for the original series of meetings
• [x] We need to generate backup meeting links for recording purposes as Zoom accounts only allow co-host privilege to people of the same organisation. UCL and/or Erasmus to create alternative Zoom/team links to host, in case main link cannot be used for recording. (@dmitrij176 @mattodd @MA-Jjingyi @wwjvdsande)

List of Epichem compounds to address the SAR gap

Hung has made his list of potential compounds for closing the SAR gap. @dmitrij176 to independently look through the epichem library to find compounds for SAR gap. We will merge the two list together, then ask Martine to ship the compounds in one shipment. In meeting:

Series 1 Manuscript Draft Hung has updated chemical data and SAR onto the draft. Hung also updated chemical data on the in vivo document. In meeting: Mat and Wendy are waiting for Peter Rutledge and Alice to finish editing the current draft before doing further edits. The preferred way of editing the draft is to use Dropbox Desktop, not Online. i.e. open the Word version on a computer and upload/sync it with the online version.

• [ ] @dmitrij176 to complete his inputs to the experimental section
• [ ] @fantasy121 and @dmitrij176 to assemble the NMR spectra (images of spectra, and raw data files) for novel compounds - location needs to be decided.

Screening compound series in vitro @MA-Jjingyi screened compounds from series 1 and series 6 in vivo as well as some of the natural products. So far only one fenarimol significantly prolonged larval survival. No activity was seen in the others tested. Also no activity was seen in the ketoximes. When the triplicate data is available the survival data will also be added in the spreadsheet.

• [ ] @dmitrij176 to summarise (in a new post, with a link to that post from the wiki) latest in vitro and in vivo data from Aug and Sept in advance of the next meeting.

RACI2022 Hung shared his experience at RACI 2022 conference. Fenarimols Series 1 work was presented at this conference. Rohan Davis (Mat’s OSM collaborator @mattodd) was interested in sending some anti-malarial compounds from his lab in Griffith University for mycetoma testing. (need to post structures)

• [ ] @fantasy121 to contact Rohan for follow-up, to obtain structures and whether they are intended for a particular biological target.

Series 2 and other series

• [x] @kym834 to update on series 2 work with SSP and high school students and double check there is no doubling of compounds synthesis from different participating high schools (Australia vs Seven Oaks for eg). In meeting: An update on which molecules have been made, and which are planned, as well as an updated synthetic scheme, was needed from someone on the Breaking Good team @KlementineJBS @alintheopen since there was nervousness from e.g. Sevenoaks team about unintentional duplication of effort.

@AndreaOtago has sent 17 compounds which are currently prepared for screening @csuth22 will sent additional compounds as well. Codes need to be assigned to these compounds and the Master List needs to be updated. Can we please have a volunteer? In meeting: @fantasy121 agreed to do this.

• [ ] @fantasy121 to check the compounds in OpenSourceMycetoma/series-5-benzimidazoles#3 and ensure these compounds had been assigned MyOS Codes.

Updates:

• USYD. In meeting: @fantasy121 shared a set of ca. 10 targets he is currently exploring synthetically. @bebi78 asked about metal complexes of these compounds.
• [ ] @fantasy121 to share that scheme below.
• UCL. In meeting: @dmitrij176 is moving towards further ketoxime analogs and a potential separation of the diasteroisomers.
• Erasmus. In meeting: @MA-Jjingyi shared new in vivo data as mentioned above and in the slide deck on this page.

1. Have we captured all the recent samples contributed by @bebi78 ? This is not clear
   • [ ] @bebi78 to advise whether all the samples he has contributed are in the Master List already. A presentation was mentioned in the last meeting from @MMos2022 - what was this, and is it still needed?
   • [ ] @wwjvdsande to chase this and post below. Was there a recording for the last meeting? In meeting: no.

In meeting: @wwjvdsande reported that she had secured a grant that will allow her to continue screening for another 18 months, which is great news.
