O59757 Spc7 SPCC1020.02 phosphorylated form PMID:22521786
residues T453 and T507 (simultaneously) (and a generic "unphosphorylated" form)

nataled commented 2 years ago

Hi Val, For clarification, by 'generic unphosphorylated' do you mean a term asserted to have no phosphorylations anywhere on the molecule, or just for the indicated positions? Based on the statement in the paper "mutation of all nine threonine residues of the MELT motifs (Spc7-9TA) reduced Mph1-dependent phosphorylation of Spc7 by 63.5%" I surmise that a true fully unphosphorylated form is unwarranted. Please advise.

Actually it should be Spc7 P-MELT (T257, T338, T366, T395, T422, T453, T507, T529, T552) (metaphase, although in reality more residues are phosphorylated by Mph1 in metaphase I don't know which ones)

this paper can be the reference for the phosphorylated form

I know that the phosphorylation is removed at anaphase, but I don't know which residues. are removed. Spc7 is a target of 3 kinases , but I think only one phosphatase removes all of the P sites (the phosphatase repertoire is much smaller and dis2 antagonizes all these 3 kinases).

Let's leave the un-P form for now. I will just model the metaphase version as unknown phosphorylation state

nataled commented 2 years ago

Sorry, I'm now a bit confused about the phosphorylated form. Are you requesting the original two-position phosphorylated form, or are you requesting a nine-position phosphorylated form?

ValWood commented 2 years ago

Sorry, scrub the 2-site. one. I'll get the 9-site one as this is likely more representative- and represents the version in this paper.
