Inspiration from "Pandemic" the board game

Each player may select an ability/class right at the start of the game. He will keep this ability throughout the game.

Engineer: Increase level of 1 tower for 1 wave, cannot be used on the same tower for X rounds

Magician: May swap the position of two towers permanently, recharges each wave

Scientist Overclock: Buff one kind of tower for one wave, the next wave this kind of Towers is debuffed

Trump: May build up to 5 wall pieces that can be build an demolished for free

Gambler May designate one tower, all kills made by this tower give extra gold

Broker: Disable a turret for one wave you get 10% of its cost after the wave

Some more general ideas:

Different Player Powers

Inspiration from Root

Each player has a distinct gameplay style. For example, one player constructs towers, another gathers resources (such as mana) to cast spells (buffs, debuffs), and a third player builds and upgrades troops to combat creeps. One player can even take on the role of the creep overlord, spawning and upgrading creep waves.

Inspiration from Cosmic

Each player possesses a unique power that significantly influences their gameplay style.

Tower Placement Restrictions

Inspiration from Blood Rage / Rogue Tower

Players receive random upgrade cards or entirely new and unique tower types, leading to diverse play styles. These upgrade cards could also be unique to each player, further differentiating their strategies.

Inspiration from Exodus Proxima Centauri

Towers feature slots for upgrade items or modules. Players acquire these upgrade items either randomly or by selecting from a limited selection.

Inspiration from Element TD

Each player can build only a subset of all available towers in a game. For instance, one player might focus on dealing more type X damage, another on area-of-effect damage, and another on damage over time, freezing, stunning, or applying debuffs/buffs.

Synergies

Certain towers gain special bonus effects when placed near each other, or players must arrange their towers in specific patterns to achieve combos. These combos require cooperation, as no single player can achieve them on their own.
