Peristaltic pump works WAY better. No doubt about it. I don't have the 5um top filters yet so I just filtered seawater with an algal density I would expect to have in my experiment through the 0.7um and then the 0.22um and it worked really well. No reason to believe a top filter would impede flow rate.
Test 3 inline Swinnex filter apparatuses (~10um, 0.7um, 0.22um) to capture ciliate, phytoplankton, bacteria, using peristaltic pump to move water through.
You can use any seawater. This is system we decided (Brook, Brent, Emma) would be most efficient (time saver) and give us proper samples.
