Thanks, group Zinc! Here is some points of feedback on your written proposal. Your TA mentor will also provide feedback and the results of marking.

• why would genetically-induce T2D be on the rise?
• age/sex are included in the hypothesis, but no rationale is given
• "genetic compositions" implies DNA differences, but I think your data only allow you to survey transcriptome differences
• what is 'microarray RNA sequencing'?
• key details about how many samples per combination of T2D/healthy and Obese/non-obese, as well as per sex are missing - these details are necessary to evaluate whether the hypotheses can be addressed with this data
• how does the single-cell data help address the hypotheses, which are all seemingly at the patient level?
• for aim 2, is cell type considered? It is not clear what a t-test post DESeq2 is doing.
• I suggest to explore alternative approaches for DE testing in single cell RNAseq (lectures 14-15 will be useful here!)

Here are some of my feedback:

Question / Hypothesis: I agree with Keegan here. There is no explanation on why you are considering sex and age as variables here too. Do you expect to see different gene expression signatures between these variables?

Data: Missing the data distribution 9T2D and Obesity - also if you are considering age and sex that too)

Methodology: This could be more detailed. HOw are you planning to compare your results with the original study? Is there any downstream analysis you would want to do to elucidate the relationship between T2D and Obesity?
