Analysis and explanation. The gene ENSG00000138109 encodes for a cytochrome P450 monooxygenase, which metabolizes various endogenous substrates such as fatty acids and steroids. It catalyzes reactions involving the insertion of an oxygen atom into substrates and the reduction of the second oxygen atom into a water molecule, using electrons from NADPH via cytochrome P450 reductase. The gene's documented catalytic activities include epoxidation of polyunsaturated fatty acids, hydroxylation of carbon-hydrogen bonds, metabolism of cholesterol into 25-hydroxycholesterol, formation of catechol estrogens, hydroxylation of fatty acids and metabolites of plant monoterpenes, and metabolic activities related to drugs like S-warfarin and diclofenac. The provided reaction, which converts testosterone to estradiol-17beta, formate, and water, does not align with the described functions and substrate specificities of this gene, indicating that it is not relevant to the reaction. No

Analysis and explanation: The gene ENSG00000186377 encodes a cytochrome P450 monooxygenase that specifically catalyzes the epoxidation of anandamide and does not exhibit hydroxylase activity towards fatty acids, steroids, and prostaglandins. The reaction described involves the conversion of testosterone to 19-hydroxytestosterone, a process that the enzyme encoded by this gene does not participate in, as its catalytic activity is specific to the epoxidation of anandamide and not the hydroxylation of testosterone. No

Analysis and explanation: The gene ENSG00000186160 encodes a cytochrome P450 monooxygenase that primarily catalyzes the in-chain oxidation of fatty acids, hydroxylates lauric and myristic acids, and catalyzes the epoxidation of double bonds of polyunsaturated fatty acids, using NADPH via cytochrome P450 reductase. The annotated catalytic activities include hydroxylation of fatty acids like dodecanoate and tetradecanoate and the epoxidation of arachidonic acid. However, the provided reaction involves the hydroxylation of testosterone to produce 19-hydroxytestosterone, which is not listed among the catalyzed reactions for the referenced gene. Therefore, there is no direct support connecting the gene's function to this specific reaction involving testosterone. No

Analysis and explanation. The gene ENSG00000108242 codes for a cytochrome P450 monooxygenase involved in retinoid metabolism, specifically hydroxylating all-trans-retinoic acid (atRA) to 4-hydroxyretinoate. This process involves molecular oxygen and NADPH. However, the reaction provided involves the conversion of testosterone to 19-hydroxytestosterone, which is unrelated to the functionality of the cytochrome P450 monooxygenase described for ENSG00000108242. Many of the reactions involve cytochrome P450s, which can often can take quite a few different substrates, so that a very thorough curation would be required to further clean this up. Also there seems to be some reactions localized to the cytosol while others are in ER.

For a few reactions it was not too difficult to resolve what should be the new gene associations:
