When downloading it that way, you need to choose an E-ink Kindle (not a Fire) and enter its serial number into the plugin customization.
Pardon my ignorance, but what’s an E-ink Kindle? It sounds like some kind of device. I don’t have a Kindle device; I use the Kindle app on my iPhone.
I have a Kindle book in .azw3 format on my Windows 10 computer, that I downloaded from my Amazon account using the “Download and transfer via USB” function.

I don't understand how you were able to do this.
The "Download & transfer via USB" function only allows you to select a Kindle e-reader device or Fire tablet as the intended target of the download. It will not allow you to select the Kindle app on a phone. Unless perhaps you have the old Fire phone, but if so it won't work with DeDRM. (Or perhaps there is some other Kindle phone that I am not aware of.)
If you don't have an actual Kindle device then you will need to use an old version of Kindle for PC or Kindle for Mac to obtain a file that can be used with this software.
@j-howell Which is the most recent Kindle for PC version compatible? I went way back and I still can't import them properly.
You can use any version prior to 1.25. See this thread on MobileRead for details of how to get different versions to work.

When I try to convert the book to .pdf using Calibre, even with the DeDRM removal plugin installed, it still says it can’t convert it because of DRM. I deleted the book from Calibre and re-imported it a couple of times, but I still get the same result.
