In Firefox's "Cookie Banner Handling/Blocker" setting, ImprovedTube's "Up Next Autoplay -> Off" feature cannot implement.
Re-produce steps for the issue:
1) Download Firefox (any release). Or if the installed, run Firefox with a new profile on Command Prompt/Terminal:

2) Go to about:config.
3) Search cookiebanners.service.mode setting. Change as 2 or 1.
4) Install ImprovedTube add-on.
5) Open the ImproveTube settings. Turn off "Up Next Autoplay" in Player.
6) Open a Youtube video. For example.
Result: Autoplay toggle button ('Autoplay is on') still is enabled on player control. Naturally, when the video is finished, the countdown starts for the next video play.

Alternative solution 1: Install Disable YouTube Autoplay userscript.
Alternative solution 2: Install Enhancer for YouTube. Of course, need select to "Disable autoplay" from the settings of the add-on.
