Hi, Is there a way to extract species-specific kmers from the database and assemble them into species-specific sequences as long as possible? These can then be used to design diagnostic PCR primers.

This idea is interesting, it can output species-specific kmers, but only for one end. For the other end, the code would need to be customized and modified according to your idea.
