I have multichannel samples and need to obtain deconvolved single channel tiff stacks. While deconvolution of one channel works fine using a bead-extracted PSF from this channel, is fails to extract a PSF from another channel. Beads here are very dim. I work around this by using the PSF extracted form the first channel. This results in visually satisfying results but is reducing the potential quality of the deconvolved data.
