nterPro:IPR001830 trehalose related #309

Hi Sandra,

I queried the annotation of TPS2 with SGD and it appears this is correct.

Could you therefore check the mapping to "alpha,alpha-trehalose-phosphate synthase (UDP-forming) activity" should not be trehalose-phosphatase activity (it seems the complex has both activities?)

cheers

Val

Val,

The annotation we have for TPS2 (trehalose-phosphatase activity) is correct. TPS2 is similar to TPS1 which has alpha,alpha-trehalose-phosphate synthase (UDP-forming) activity. I am guessing Interpro picked it up because of the sequence similarity.

The following paper clearly identifies the role of TPS2 as a trehalose 6-phosphate phosphatase (E.C-3.1.3.12). De Virgilio C, et al. (1993) Disruption of TPS2, the gene encoding the 100-kDa subunit of the trehalose-6-phosphate synthase/phosphatase complex in Saccharomyces cerevisiae, causes accumulation of trehalose-6-phosphate and loss of trehalose-6-phosphate phosphatase activity. Eur J Biochem 212(2):315.

You may also want to check the trehalose biosynthesis pathway page to see the role of TPS1 and TPS2 in this pathway. http://pathway.yeastgenome.org:8555/YEAST/new-image?type=PATHWAY&object=TRESYN-PWY&detail-level=2

Please let me know if you have any questions.

Best,

Rama

I've taken the function mapping up to catalytic activity for this one, looks like the domain is in too broad a span of enzymes to do anything else.
