I'm not sure of the 'directed killing' part of your definition...do you think your defs work well for toxins.. Not sure of cytotoxicity being = to cytolysis either.. I think cytotoxicity can occur without eventually causing cytolysis...what do you think?

Hello, I have to agree with Evelyn that I don't think "cytolysis" = "cytotoxicity". Also, it seems that the spur to changing the definition of this term in GO is because people in the T-cell, NK cell, and ADCC communities are inappropriately calling their process "cytolysis". In my experience "cytolysis" always refers to a rupture of membranes. I agree that GO needs a "cell killing" term of which "cytolysis" should be a child. I suggested one back in the winter - that whole thread has been put on hold until the next content meeting in August since there seemed to be difficulty in reaching consensus (over the pathogenesis issue). In addition, there is an effort underway by a group called PAMGO that will be suggesting some new "cell killing" terms to the GO in the very near future. Perhaps a generic "cell killing" term is the better way to go rather than redefining something which, I think, has a pretty clear biological meaning. Just my two cents... Michelle

I'm quite happy to shift the cytotoxicity terms to be children of a new immune cell mediated cytotoxicity term, rather than to be children of cytolysis. In fact that's how I originally wrote up the entry. I switched to having these terms under cytolysis because somebody earlier had placed natural killer mediated cytolysis under cytolysis. This was not a surprise to me, because immunologists had long used the term cytolysis as a synonym for NK and T cell killing of target cells, due to an earlier misunderstanding of the mechanisms involved. When I started in immunology, it was thought that perforin release from these cell types literally lysed the target cells, although now it is known that perforin acts in physiological settings only to facilitate the entrance of granzymes into the target cells. I assumed therefore that cytolysis was being used in the GO with a broader meaning, and thought I should bring the definition in line with that.
Anyway, it makes sense to separate T-cell mediated cytotoxicity, NK cell mediated cytotoxicity, and ADCC, all of which share common elements, away from cytolysis, which then can be reserved for nasty microbial gene products and perhaps C9 and other members of the complement cascade.
Regarding the "directed killing" part of my definitions, my intention is for these terms to be used to describe contact- dependent killing of target cells by immune system cells (in many or most situations following the formation of an immunological synapse), not for the random killing of susceptible cells following the release of cytokines in the broader environment.
Since I do not then wish to change the GO definition of cytolysis, and want remove the immune system-related terms from being its children, I do not believe that this entry then bears on the issue of pathogenesis or toxins and therefore the terms can be entered in the near term. We really need a "T-cell mediated cytotoxicity" term in the GO, as this phenomenon is central to immune responses to virally and bacterially infected cells. Also, at the moment I do not think an additional "cell killing" term is needed in the GO, since GO:0008219 cell death already exists as grouping term.
Thus, I have made appropriate changes below.

The problem with the original definition is that, despite its etymological formation, the word cytolysis has been widely used to describe any type of killing of target cells, whether through apoptotic or non-apoptotic mechanisms. Indeed, it is now clear that T-cell killing of target cells through perforin dependent mechanisms, NK cell killing, and ADCC are all apoptotic mechanisms in physiological circumstances, yet these are still termed cytolysis, even though it is clear that large scale membrane rupture is not part of the process, and the loss of cytoplasm is through cellular blebbing.
Hemolysis is non-apoptotic and as defined in the GO, a lytic destruction of another organism's erythrocytes.
Thus, I believe the best way to capture the de facto meaning of cytolysis is to define it as essentially any means of killing target cells (whether self or non-self), and use other process and function terms to point to mechanistic features of particular processes.
