new terms: cytosine methylation, adenine m... #3720

Hello,

I'd like to request 2 new terms.

% DNA methylation (GO:0006306) %% adenine methylation (NEW) %% cytosine methylation (NEW)

DEF: The covalent transfer of a methyl group to N-6 of adenine in a DNA molecule. DEF: The covalent transfer of a methyl group to C-5 or N-4 of cytosine in a DNA molecule.

Hi,

It might be worth checking if it's always those positions that get methylated, even in other organisms, kingdoms etc. I seem to recall that E. coli has DNA methylation too, so if the terms with names that are just 'adenine methylation' specify position within the base, then we need to make sure it's universal. Otherwise, we might need more terms, i.e.:

--DNA methylation (GO:0006306) ----adenine methylation (NEW) ------adenine N6 methylation (NEW) ----cytosine methylation (NEW)

etc.

EC has enzymes that methylate adenine N1 or N6, cytosine C5 or N4, and guanine N1, N2 or N7, but the guanine methylations and adenine N1 methylation occur in tRNA, not DNA. (Also, all of the DNA methylases available from NEB are adenine N6, cytosine C5, or cytosine N4, and most of them are bacterial.) So I think the defs would be OK.

I've added the terms as

- DNA methylation on adenine GO:0032775

... to avoid confusion with methylation of free adenine or cytosine ('adenine methylation' and 'cytosine methylation' are synonyms).
