regulation of response to tumor necrosis factor #5454

Hi

Today I am annotating ADIPOQ Q15848 with PMID:10604883 data.

Treatment with ADIPOQ before stimulation with TNF reduces the TNF enhanced adhesion of monocytes to aortic endothelial cells.

So I was wondering if the NTR: negative regulation of response to tumor necrosis factor could be created.

Do you think it might be better to use the (not yet existing) term 'negative regulation of tumor necrosis factor-mediated signaling pathway'?

The signaling pathway is the response to TNF. It's a more specific and descriptive term.

We may even want to merge 'response to TNF' and 'tumor necrosis factor-mediated signaling pathway' because we can't think of differences between these two. Can you?

I think it is necessary to get the 'response to' terms with more specific definitions so that people are clear about what can be associated with these terms (see response to SF items/reference genome). However having a quick browse of the response to ontologies I wonder if something more constructive could be sorted out.

Basically if you delete response to TNF then I guess you would need to delete loads of response to terms which are in a similar vein, response to insulin, hormone, cytokine, pheromone.

Interestingly response to hormone stimulus has the child terms GO:0009755 hormone-mediated signaling GO:0043434 response to peptide hormone stimulus GO:0009720 detection of hormone stimulus GO:0048545 response to steroid hormone stimulus etc and I wondered if actually the signaling pathway terms should be child terms of the response to hormone/cytokine etc stimulus?

Although if it is generally agreed to cut down many of the response to terms in this way (by incorporating them in signaling pathways) then I agree I personally think this is probably what is meant by response to ..... but at present there are wide variations in how to interpret the start/stop of the response to processes.

In the meantime, if you create negative regulation of tumor necrosis factor-mediated signaling pathway I would be happy to use it!
