Factor X activation #6012

Hi

please could the following new term be created:

GO:0002541 activation of plasma proteins during acute inflammatory response > is_a child New Term: Factor X activation

GO:0002543 activation of blood coagulation via clotting cascade > part_of child New Term: Factor X activation

Any process that activates Factor X. Factor X is a protein synthesized by the liver that circulates in an inactive form until excision of the tripeptide RKR to produce the active two-chain Factor Xa. Mature Factor Xa can subsequently cleave protein substrates (converting prothrombin to thrombin). Factor X is a participant in the clotting cascade (coagulation cascade) and is the first member of the final common pathway or thrombin pathway.

As phrased this is a request for a process term to describe a single, specific protease (factor III) acting on its specific substrate (factor X). It's an instance of the more general phenomenon of amplification in a signaling cascade mediated by proteolytic activation.

Hi

I requested this based on the existence of GO:0002542 Factor XII activation.

The advantage of this term is that it links it's parent terms: GO:0002541 activation of plasma proteins during acute inflammatory response GO:0002353 plasma kallikrein-kinin cascade

Similarly the term Factor X activation would link: GO:0002541 activation of plasma proteins during acute inflammatory response GO:0002543 activation of blood coagulation via clotting cascade

But I agree this is a single enzyme step, and potentially this would lead to a request for a term for the activation of each plasma factor, but presumably these would link to multiple parents. I guess this is more like an extension of the idea of having a function as part of a process term!

I look forward to your thoughts on this

Actually, the representation of 'factor XII activation' is incorrect since the activation involves a conformational change and not proteolysis. It should be represented as a positive regulation of the proteolytic process. We moved this term to be an is_a 'positive regulation of proteolysis' and we made it positively-regulate the 'activation of plasma proteins ...' term.

In the long run, we will probably obsolete this term and make a process-specific function since it describes a single molecular event. We will also represent the factor X term this way too.
