Q about ergosterol biosynthsis #6698

GO:0007009 plasma membrane organization --GO:0031579 membrane raft organization (is not a child, is there any reason, are there membrane rafts which are not plasma membrane?) ----GO:0008204 ergosterol biosynthetic process

As ergosterol is reponsible for membrane permeability and fluidity?

We've covered this before; yes, there are membrane rafts that aren't in the plasma membrane. I'll add a term (probably tomorrow) for localization-to-raft, but it won't go under loc to plasma membrane.

Organization doesn't include biosynthesis, so I wouldn't add ergosterol biosynth part_of raft organization. I also wouldn't make the existing ergosterol synth term part_of a raft biogenesis term, because ergosterol is also a precursor of vitamin D -- nothing to do with membranes there!

added protein insertion into membrane raft GO:0071210 (used that name to match existing terms; 'establishment of protein localization to membrane raft' is exact synonym)

I'm a bit confused by the history of the membrane raft stuff. Do we have an example of a a structure described as a "membrane raft" that isn't assiciated with the plasma membrane? I could't find one from scanning the old threads but i might have missed it.

We based the GO parentage on this paragraph from the paper cited for the revised def (PMID: 16645198; full text http://www.jlr.org/cgi/content/full/47/7/1597):

'Although "plasma membrane" was suggested for inclusion in the definition of membrane rafts, it was quickly excluded from consideration. Presentations and posters indicating the existence of raft-like domains on intracellular membranes such as the endoplasmic reticulum and mitochondria made it clear that the plasma membrane does not hold a monopoly on membrane domains. Although the nature of such "non-plasma membrane" membrane domains is not yet known, room must be left in the tent to welcome these newcomers.'

I did find one paper on raft-like ER membrane domains - PMID: 16835267; http://jcs.biologists.org/cgi/content/full/119/15/3149. And PMID: 12208858 (http://www.jci.org/articles/view/16390/version/1\#B3) says rafts assemble in the ER in mammalian cells.
