Hi Yasmin,

I found the following term in GO:

GO:0043491, protein kinase B signaling EXACT SYNONYM: AKT signaling cascade Definition: A series of reactions, mediated by the intracellular serine/threonine kinase protein kinase B, which occurs as a result of a single trigger reaction or compound.

Could that be what you are seeking?

If your process is different, can you provide a definition for the signaling pathway please? You can consult with Becky on the current structure of the signaling branch of the ontology and how such terms are currently defined so that the new term will be consistent with the existing ones. Also, placement within the existing structure would be great.

You know, I think I will reassign this item to Becky so that she can give it the full treatment as she has much more background in the signaling area of GO than I do.

I think the PI3K/Akt signaling pathway is different to that of GO:0043491 protein kinase B signaling, as this one is a retinoid-activated pathway involving the lipid kinase phosphatidylinositol 3-kinase (PI3K). There is detailed information about this pathway at http://rgd.mcw.edu/wg/pathway/phosphatidylinositol\_3-kinase-akt\_signaling\_pathway. [term]: Phosphatidylinositol 3-kinase-Akt signaling pathway [definition]: Phosphatidylinositol 3-kinase-Akt signaling pathway plays a crucial role in many important cellular processes. PI3K-Akt pathway is one of the two conduits of insulin signaling. Also, PI3K-Akt pathway, possibly via Akt2, is believed to play a role in the insulin responsive glucose transport. PI3K-Akt plays a central role in activating mTOR pathway by removing the inhibitory effect exerted upon the mTorc1 complex of mTOR. mTOR signaling controls protein translation and ribosome biogenesis, among others. Hi Yasmin and Tanya,

AKT and PKB (protein kinase B) are the same thing. In many pathways (eg insulin receptor signaling), PI3K acts upstream of AKT/PKB to activate it. AKT/PKB can also be activated by PI3K-independent routes too though.

I agree with Tanya that the best existing terms to annotate to are GO:0043491, protein kinase B signaling and also to GO:0014065 phosphoinositide 3-kinase cascade (or their regulation children)

I'll have a think about how we can link up the two (PI3K and PKB/AKT that is).

We discussed in the signaling group adding in terms to connect the intracellular signaling cassettes to the upstream receptors, so I can add terms for 'PKB pathway involved in retinoic acid receptor signaling'. We're still resolving the exact wording of these terms so I'll take a proper look after the GO camp.
