Hi Ev,

Looking at the existing terms, I think the new term should be named 'tolerance induction to endotoxin' and be an is_a child of 'tolerance induction to nonself antigen' (GO:0002462). OK?

Evelyn and Midori

Three things:

1) I would prefer the term name be "tolerance induction to lipopolysaccharide", since occasionally endotoxin is used with a broader meaning than LPS, which at least has a general meaning in regards to its structure. Thus "tolerance induction to endotoxin" could be a broad synonym, and "tolerance induction to LPS" an exact thing.

This naming is consistent with all other LPS terms in the GO.

2) I agree with Evelyn's initial placemant of the new term under "tolerance induction". LPS tolerance induction occurs within individual cells that have been over-exposed to LPS and involves internal mechanisms to the cell that modify the functioning of the LPS (TLR4) signaling pathway. This does not satisfy the definition of "'tolerance induction to nonself antigen'" because it does not satisfy the definition of its parent "tolerance induction dependent upon immune response" since it is not dependent upon a T or B cell response and does not involve the mechanisms of anergy, cellular deletion, or regulatory T cell activation.

3) The new term could also have "cellular response to lipopolysaccharide", "negative regulation of lipopolysaccharide-mediated signaling pathway", and "negative regulation of toll-like receptor 4 signaling pathway" as parents.

It's good to have many parents, isn't it?

Thanks,

Alex

Hi Alex,

Thanks for your recommendations -- I can always count on you for much deeper insight into these immunology requests than I can bring!
