Hello,

could you create new children of GO:0048010 vascular endothelial growth factor receptor signaling pathway? This would greatly clarify matters, since there are several receptors for VEGF family members, and their functions are non-identical (see the recent Reviews PMID:22762016 and 21711246)

a) FLT1 signaling pathway (FLT1=VEGFR1) A series of molecular signals that starts with the binding of VEGFA, VEGFB or PGF to the tyrosine kinase receptor FLT1 on the surface of a cell, and ends with regulation of a downstream cellular process, e.g. transcription. VEGFA, VEGFB or PGF binding to the receptor FLT1 mediates receptor dimerization, activation of its intrinsic tyrosine kinase activity and autophosphorylation. The activated receptor then phosphorylates various substrates, thereby activating distinct signaling cascades within the cell that trigger a change in state or activity of the cell.

b) KDR signaling pathway (KDR=VEGFR2) A series of molecular signals that starts with the binding of VEGFA, VEGFC or VEGFD to the tyrosine kinase receptor KDR on the surface of a cell, and ends with regulation of a downstream cellular process, e.g. transcription. VEGFA, VEGFC or VEGFD binding to the receptor KDR mediates receptor dimerization, activation of its intrinsic tyrosine kinase activity and autophosphorylation. The activated receptor then phosphorylates various substrates, thereby activating distinct signaling cascades within the cell that trigger a change in state or activity of the cell.

c) FLT4 signaling pathway (FLT4=VEGFR3) A series of molecular signals that starts with the binding of VEGFC or VEGFD to the tyrosine kinase receptor FLT4 on the surface of a cell, and ends with regulation of a downstream cellular process, e.g. transcription. Binding of VEGFC or VEGFD to the receptor FLT4 mediates receptor dimerization, activation of its intrinsic tyrosine kinase activity and autophosphorylation. The activated receptor then phosphorylates various substrates, thereby activating distinct signaling cascades within the cell that trigger a change in state or activity of the cell.

Many thanks in advance,

Ursula

Hi Ursula,

I used the protein ontology for the names, but kept your suggestions as synonyms.

vascular endothelial growth factor receptor 1 signaling pathway ; GO:0036323 vascular endothelial growth factor receptor 2 signaling pathway ; GO:0036324 vascular endothelial growth factor receptor 3 signaling pathway ; GO:0036325

Also added more specific receptor terms: VEGF-A-activated receptor activity ; GO:0036326 VEGF-B-activated receptor activity ; GO:0036327 VEGF-C-activated receptor activity ; GO:0036328 VEGF-D-activated receptor activity ; GO:0036329 VEGF-E-activated receptor activity ; GO:0036330

Thanks, Becky
