1) Currently the POG program maintains their own internal knowledgebase of variant interpretations that they have developed throughout the course of the trial. We will merge this knowledgebase with CIViC and integrate CIViC’s API into the POG analysis pipeline. We have already incorporated 114 POG clinical interpretations into CIViC. However, ~90% of the POG knowledgebase is comprised of variants with only evidence of oncogenic/biological function. With the expansion of CIViC’s data model in #5 these too will be merged into CIViC. The resulting knowledgebase will then be used to generate interpretive reports for both retrospective and prospective POG cases. 2) To measure the impact of CIViC interpretations on decision making process, we will conduct a survey-based analysis on a groups of physicians who use the CIViC database to inform treatment decisions for oncology patients with genomic sequencing data. The POG group has created a molecular tumor board (MTB) to discuss appropriate steps for patients undergoing treatment after sequencing analysis. We will conduct surveys on each case that undergoes MTB review. 3) These surveys will be collected using a HIPAA compliant database to ensure protection of all identifiable information in accordance with the IRB at the Washington University School of Medicine. Conducting this analysis will require amendment of an existing IRB obtained from the Washington University School of Medicine (HRPO #: 201801112) and filing for a new IRB with the BC Cancer Research Ethics Board.
