Team 1: Hardware design

We will work on designing an hardware and the membrane in it. We will try to approach and answer the following points:

• Adherence to a membrane
• Design of the hardware and the membrane
• Pre-sorting the minicells by size, filter them with the membrane and purify them (with team 2) flow and movement
• What separation method we have to use
• Looking at the “mother machine” designed at the CRI

1) How can we isolate minicells from mother cells and the cell component from dead mother cells Double filter design:

• first filter: let the minicells pass-through
• second filter: don't let the minicells pass-through The minicells will be stuck between these two filtrations systems.

2) What kind of hardware? Our first idea was a microfluidic system - based on the idea of the mother machine but with a core channel and large sides for the minicells to be "stored". We also thought of a more low-cost system in a pretri dish. We would have a 45°C to allow the minicells to be directed through the filter. These ideas are still under construction

3) What kind of membrane to attach the mother cells? We want to have a membrane to which the minicells will be attached -without genetically modifying the mother cells for this. One issue of this part is that the minicells would therefore also potentially be attached to it.
