RBS and BCD collection #134

Open traci-igem opened 2 years ago

I'm adding in this issue more as a note to myself, but also open to ideas - we need a collection of bacterial RBS and BCDs for the distribution. Please toss out ideas / suggestions / etc for these parts. I'll work on generating the files for the collection after the dust settles from the Jamboree.

I already created a new branch and deleted the old one. I will make a pull request with a tentative list this weekend.

In addition, it could be very useful to build a standard collection that enables the sub-assembly of new iGEM-assembly-compatible BCD parts, from standard RBS parts and a collection of leader peptides. Some cleverness will be required in the design as the leader peptide parts will eventually need to expose the same overhang on their 3' end (AATG, for pairing with the CDSs) as was initially exposed on their 5' end (AATG, for pairing with RBSs). This could be accomplished by using a different IIS restriction enzyme to expose the 3' AATG overhang, or by using a conditionally methylatable BsaI (mBsaI) cut site (see a paper on this here, and an example design of linkers that use mBsaI sites to expose new overhangs here).
