On the Settings tab, press detect on Red Dead Redemption 2. Do you see a window asking to rename the folder?
Also, I see that you have both folders on Program Files, you need to run the program as an administrator.

Even though I'm running it as admin, and Windows 10 Dev Mode is enabled, I get this error now

Try to close all of the launchers for RDR2 (Rockstar Games, Epic Games and/or Steam) before clicking detect.
If that does not works, follow this steps:
C:\Program Files\Rockstar Games
, rename Red Dead Redemption 2
to Red Dead Redemption 2 - Clean
C:\Program Files\Rockstar Games\Red Dead Redemption 2
