It doesn't work if you have a pin code enabled.
Zune Desktop reports that it cannot talk to your phone if the screen is currently locked, but works after you unlock - libmtp-zune simply doesn't work if you have a lock enabled, even if not currently locked.

I'm trying to get signs of life from my Lumia 800 WP7 device.
I'm not seeing any evidence that the MTPZ handshake is even being attempted, even though the microsoft.com/MTPZ string is definitely being received from the device:
