Is Amstrad PPC 640 3.5” floppy drive supported? I’ve read that it’s Shugart interface instead of IBM and pinout is a bit different.

Reply for those who will search: yes, supported. When you tell greaseweazle --drive=0 , it will will recognize it as Shugart-34 interface.
