Change vignette to use mutate for smooth & deriv #84

This enables easier use of the blank argument & is probably just better practice (even though subset_by exists)

Also this creates a more natural emphasis of the fact you want to be using unique well in each plate for smoothing and derivs

AKA plate-well is not equivalent to just well
