The intensity of the excitation light increases toward one side as the LEDs on the LED board headed in one direction. The simple trick is putting a black grid between the low heater board and the LED board to isolate individual wells like this photo. 20200925_180039

Another idea - albeit requiring more extensive design changes - would be to illuminate individual wells sequentially so that the cross-talk is eliminated by spreading the illumination and sensing from each well in time.
