I suppose the special "slots" would be the way to go. ELR works quite simply. You have a recipe and click on the button where you can also "Edit" or "Adapt" that recipe for example. You then choose "Make One-Shot" and you can put in how much of the Oneshot you want to make - and the website will provide you with information on what mixing percentage you should use it when mixing with base, how much you need of each flavour, as well as how much you'll be able to make with it. If you want to calculate the base and nicotine shot, you have to create a whole new recipe with one flavour, that is the one-shot Something like a "Mix Oneshot" button, where you simply put in the concentration of that would be nice.
I don't use that, so I don't know how complicated it is to do that in ELR. I know the calculator I use http://diyjuicecalculator.com/ has that feature. Maybe check that out for inspiration?
Open: how do we want to track the one-shot once it's created? We'd probably don't want to add it to our flavor DB, so the user should have some special "slots" in his inventory / flavor stash for this?
