Very few changes or comments on the Discussion as it stands, but I think there is more material to add.
What you've done so far is a good job of disucssing the validity and issues with the work you've done. However, there is very little about the impllications of your results if they are true (or if they are false) and how it fits with the current status of the literature.
What does it mean if true? What are the implcations of, for example, Extensin splice diversity on plant development? What lessons have you learnt about the differences between G4s in plants vs animals. How might we want to think aobut these differences?
Given that you've pretty convincingly demostrated that G4s are having a regulatory effect on plant gene expression, even if you can't be certain of the mechanism, what does this mean for studies of gene expression in plants?
etc, etc. Show that you understand the biology, not just the technical details and pitfalls of data generaation and anlysi.s
adding stuff about drought response genes with G4s and tying this to possible pol II stalling, since this is probably the most relevant plant research we have built upon.
adding discussion about how truncated extensins might benefit plant development, and how this hypothesis is corroborated by current research (Baumberger et al).
Adding a final paragraph which (very briefly) summarises the implications of the research and what the implications might be for future researchers modelling gene expression or pol ii
